摘要:目的:通过分析胃腺癌的 RNA-seq和临床数据以及铁死亡相关基因数据来筛选胃腺癌潜在的治疗靶点或预后基因。方法:从癌症基因组图谱(Cancer Genome Atlas,TCGA)数据库下载RNA测序(RNA-seq)数据和临床数据,从FerrDb V2网站下载铁死亡相关基因数据。使用R软件对下载的数据进行生物信息学和统计分析,以P<0.05为差异有统计学意义。结果:我们一共筛选出1384个差异表达基因,其中包括24个与铁死亡相关的差异表达基因。在这24个基因中,我们进一步筛选了与生存预后相关的四个关键基因:NOX2、PRKAA2、FABP4和PDK4。通过单基因生存分析和Cox回归分析,我们发现NOX2、PRKAA2、FABP4和PDK4低表达的STAD患者的生存时间明显更长,差异有统计学意义(P<0.05)。结论:我们筛选出了4个与胃腺癌预后有关的铁死亡相关差异表达基因,这4个基因有可能成为胃腺癌治疗的靶点。
